What lyticase is recommended for preparation of Pichia for the PCR protocol? Can zymolase be substituted?

Answer

The lyticase for our protocol is crude lyticase. A pure preparation is more expensive and it is not necessary. We routinely use zymolase in this protocol for lysing the cells (5 ul of a 1 mg/ml stock). This is overkill, but it works fine. Ref: BioTechniques 20:980-982 June 1996.
