Peroxisome structure   Peroxisomes are single membrane–bound vesicles found in most eukaryotic cells. Their chief function is to enzymatically oxidize fatty acids and to subsequently catalyze the breakdown of H2O2, a by-product of fatty acid degradation. They are critical in maintaining the normal function of both brain and lungs in mammals and are involved in energy metabolism. Peroxisomes can be investigated in live cells using fluorescent fusion proteins targeted to the organelle, and in fixed cells using antibodies to peroxisomal membrane proteins.

CellLight® reagent for peroxisomes

Use CellLight® reagents when you want to label peroxisomes in live cells and follow the dynamics of intracellular behavior. CellLight® proteins can be multiplexed with other fluorescent probes for analysis in live cells, or after fixation for colocalization studies. These ready-to-use constructs employ the human peroxisomal C-terminal targeting sequence (0.3 kDa) to target the peroxisome with fluorescent fusion proteins. Introducing CellLight® reagents involves a simple transfection step, and they work like cell stains with minimal toxicity or chemical disruption.

Peroxisome labeling in fixed cells

The SelectFX® Alexa Fluor® 488 Peroxisome Labeling Kit provides the reagents needed to label peroxisomes in fixed cells, including cell fixative and permeabilization reagents. For detection, the kit employs a primary antibody directed against peroxisomal membrane protein 70 (PMP 70), which is a high-abundance, integral-membrane component of peroxisomes. PMP 70 is significantly induced by administration of hypolipidemic agents in parallel with peroxisome proliferation and the induction of peroxisomal fatty acid beta-oxidation enzymes.
