ChargeSwitch® Coated Plates (Direct 96 gDNA Isolation Kits)
The ChargeSwitch® Direct 96 kits are solid-phase, high-throughput genomic DNA (gDNA) purification systems designed for rapid and efficient isolation of gDNA from fresh or frozen samples. These kits utilize 96-well plates coated with the ChargeSwitch® nucleic acid-binding surface. No sample transfer is necessary with the ChargeSwitch® Direct 96 kits; sample amplification (PCR) is carried out directly in the same well as DNA binding without elution! ChargeSwitch® Direct 96 kits are easy to automate from primary sample tubes to PCR set up, without the need for expensive modules such as shakers, vacuum manifolds and gripper tools.

  • To automate the process directly from primary sample tubes, an instruments needs to be equipped with an 8 span head to access primary sample tubes as available on the Packard Janus, Tecan Freedom Evo and QIAGEN BioRobot 3000 and 8000.
  • If the process is started from a 96 well plate, the processing time can be reduced to 30 minutes with a 96 well pipetting head such as the Beckman FX and NX, the Velocity 11 Bravo, Xyril instruments or a Tecan with 96-well head.

ChargeSwitch® Magnetic Beads (ChargeSwitch® gDNA purification Kits)
The ChargeSwitch® nucleic acid purification technology is the simplest, cleanest, and most effective means of purifying nucleic acids currently available. It can be applied of the purification of both DNA or RNA and can be configured in a range of product formats including simple manual purification methods as well as high throughput automated applications. ChargeSwitch® technology is based on a unique, ionizable nucleic acid-binding ligand whose charge can be switched based on the pH of the surrounding media. ChargeSwitch® ligands have been covalently affixed to specially designed magnetic beads and combined with these beads a variety of reagents to create a line of kits for isolation of specific nucleic acids from a variety of sources.

Semi automation protocols available for magnetic bead handlers such as the Thermo KingFisher 96 and the QIAGEN BioSprint 96. Full automation on liquid handling instruments such as the Tecan Freedom Evo, Beckman Biomek, Packard Janus, Protedyne BioCube and Radius, QIAGEN BioRobots, Eppendorf ep-Motion, Velocity 11 Bravo and the Sias Theonix.

The liquid handling instrument needs to be equipped with:

GeneCatcher™ Magnetic Beads
The GeneCatcher™ technology is a novel magnetic bead-based genomic DNA (gDNA) purification technology that is designed to work on a wide range of blood samples including archived or poorly stored blood samples. The GeneCatcher™ gDNA Blood kits allow rapid and efficient extraction of genomic DNA from human blood – without the use of centrifugation. The purified DNA is suitable for use in the most sensitive downstream applications including PCR and restriction enzyme digestion. The GeneCatcher 1-3ml protocol has been automated in a 24 well format on the Tecan Freedom Evo, Hamilton Starlet and Packard Janus instruments. The instrument needs to be equipped with:

  • Ika shaker & adapter for 24 well plate
  • Gripper tool
  • Techne heating unit & adapter for 24 well plate
  • Disposable tips (1ml)

 

The GeneCatcher 3-10 ml protocol has been automated for up to 8 samples per run on the Tecan Freedom Evo. The instrument needs to be equipped with:
  • Ika shaker & adapter for 8 Falcon tubes
  • Gripper tool

Silica Membrane Filter Plates

For many years, Invitrogen has made use of silica membrane technology in the development of kits for the purification of nucleic acids for numerous applications. Kits containing the necessary reagents for single-sample preparations of either DNA or RNA are available in both spin-column format and vacuum formats. The throughput with these kits, however, is limited by microcentrifuge rotor or vacuum manifold capacity.

For higher throughput applications, Invitrogen has made use of this same silica-membrane technology in a 96-well format. Each well of the 96-well plate has the same characteristics as the single-sample kits. Invitrogen has combined this 96-well plate platform with a variety of reagents to create a line of kits for isolation of specific nucleic acids. (Product information below.)
