Choosing the correct reverse transcriptase (RT) for cDNA synthesis is critical for obtaining high yields of quality, full-length cDNA that accurately represents the input RNA. In addition, good enzyme performance at high temperatures is important for efficient reverse transcription of RNA with secondary structure, and for specificity when using gene-specific RT primers.

Reverse transcriptase enzymes with reduced RNase H activity produce more full length cDNA, and provide greater gene product representation than enzymes with wild-type levels of RNase H activity.
