FGF Basic Impacts BMP-4–Induced Differentiation

  • High biological activity
  • High purity
  • Proven compatibility with Gibco® media

Gibco® Recombinant Growth Factors are highly purified bioactive proteins. Gibco® Cytokines and Growth Factors have been bioassayed with Gibco® media. Get your cytokines and growth factors from the media company you use and trust.

In a recent publication (Cell Stem Cell 8:326 (2011)), researchers demonstrated that FGF Basic was able to alter the BMP-4–initiated differentiation of human embryonic stem cells from the extra-embryonic lineage to the mesoderm lineage.

This paper also showed evidence for the involvement of downstream signaling markers MEK and ERK and transcription factor NANOG. The implication of the work is that NANOG (and by extension FGF Basic) not only plays a role not only in maintaining the pluripotent state of stem cells, but may also play a role in lineage outcomes.

Life Technologies offers FGF Basic, BMP-4, and other stem cell–related proteins. Gibco® Cytokines and Growth Factors are high-purity recombinant proteins with high bioactivity. To help ensure GIBCO® growth factors are of the highest quality, each protein is analyzed for purity along withand structural homogeneity, to ensure so that a biologically active protein is produced.

Osteonectin Promotes Endoderm Differentiation and Cardiomyogenesis in Early Embryogenesis

  • ABfinity™ osteonectin rabbit recombinant monoclonal antibodies for sensitive detection
  • Designed for consistent performance for your assays
  • Consistent product from lot to lot

ABfinity™ recombinant rabbit monoclonal antibodies are produced from specific recombinant clones, so that antibody performance is consistent over time. ABfinity™ antibodies are available to many targets including osteonectin/SPARC and BMP-2.

Cardiomyogenesis in embryoid bodies has been shown to be driven by osteonectin/SPARC (gene ID 6678, secreted protein, acidic and rich in cysteine).

The effects of SPARC include expression of cardiac marker genes including BMP-2. During the expansion of the peripheral endoderm, thicker layers of extracellular matrix (ECM) develop, and SPARC continues to drive differentiation by regulating the molecular composition of the ECM with collagen and laminin organization in the mesenchymal parietal endoderm. ABfinity™ recombinant monoclonal antibodies are ideal reagents for signaling pathway research. The ABfinity™ technology helps ensure consistent antibody performance so you don’t have to re-optimize your assay with each new lot.

Qtracker® and CellTracker™ Reagents for Tracing Hematopoietic Stem Cells

  • Reagents provide intense, stable fluorescence that can be traced through several generations
  • Available in many colors for ease in multiplexing with other reagents and with each other
  • Excellent for long-term tracking or imaging studies of live cells, including migration, motility, morphology, and other cell function assays

Qtracker® and CellTracker™ reagents are fluorescent cell tracing dyes that are useful for live-cell studies.

These reagents provide a stable signal that can be traced through several generations, and the label is not transferred to neighboring cells in the population.

The results of a recent study aimed at understanding how metastatic prostate cancer (PCa) cells target bone

marrow hematopoietic stem cells (HSC) and gain a foothold in the organism are now available. In their experiments (J Clin Invest 121:1298 (2011)), the researchers used Qtracker® reagents for in vivo tracking of HSC and CellTracker™ reagents for in vitro tracking of osteoblast/HSC/PCa cell cocultures. Their data suggest that PCa cells directly compete with HSC in the bone marrow, which could play a role in metastasis.

Zenon® Labeling Reagents for the Study of Stem Cell Differentiation

  • Facilitates rapid (10 min reaction), one-step labeling of nuclei for FACS isolation
  • Labeled complexes are stable enough for cell sorting
  • Backgrounds are low (comparable to those associated with primary/secondary antibody approaches)
  • Ideal for labeling delicate preparations

Zenon® labeling kits were designed for simple and rapid labeling of primary antibodies. Labeling reactions accommodate 1 to 20 µg with no purification step required.

Zenon® primary antibody complexes produce low backgrounds and are stable enough for cell sorting.

Using Zenon® technology to label an antibody that recognizes an antigen present on neuronal (but not non-neuronal) brain-derived human and ape tissue, researchers were able to sort cell populations and then evaluate the methylation patterns in each group (Genome Res 21:688 (2011)). Understanding the epigenetic effects that drive the differentiation of stem cells to various fates is critical for the understanding of brain maturation and function.

When labeling delicate samples, such as isolated prefrontal cortex nuclei, speed and labeling specificity are critical factors. Antibody labeling using Zenon® labeling technology is ideal for these types of investigations.
