5' FirstChoice™ RLM-RACE for mouse CXCR4 gene and Xenopus TGF-beta-related gene.

Mouse liver total RNA and Xenopus stage 41 total RNA were treated with CIP and TAP as indicated in the RLM-RACE Kit. The treated RNA samples were ligated to a synthetic RNA adapter and used in a random-primed reverse transcription reaction. Individual PCR (A) and nested PCR (B) reactions were performed with the appropriate gene-specific primer and synthetic RNA adapter primers to amplify the 5'-end of each gene. PCR products were analyzed on a 2% agarose gel with EtBr staining. The results indicate that a product of the expected size is amplified after nested PCR. Note that CXCR4 is a moderately expressed message while the TGF-beta–related gene is an extremely rare message.
