Features include:
Binning algorithm—if discrete copy number values are not needed, copy number calls can be grouped together in bins to help increase confidence estimates. This is particularly beneficial for samples with higher copy number calls.
Multiplate analysis—multiple plates containing samples run with the same assay can be combined for copy number analysis. In specific instances, such as high-throughput analysis of many samples or assays, it is desirable to combine data from multiple plates. In some situations, combining data may improve the call rate for samples falling into copy number groups that are otherwise underrepresented on single plates.
Note: Life Technologies strongly recommends single plate copy number analysis when possible because the highest-quality copy number data is generated when measurement variation is very low.
Additional quality metrics—the ΔCt Plot view enables examination of the normal distributions of the copy number group ΔCt to discern the quality of the subdistribution clustering and separation data, and to help evaluate why a copy number call and confidence estimate was made for a given sample.
