Sensitive, target-specific quantification reagents

Based on a simple mix-and-read format, Invitrogen™ Qubit™ assays can be read in the Qubit Fluorometer following as little as two minutes' incubation. The dye in each Qubit Assay Kit emits fluorescence only when specifically bound to the target molecule, which minimizes the effects of contaminants—including degraded DNA or RNA—on the result.

Qubit Assay Kits

Qubit Assay Kits provide concentrated assay reagent, dilution buffer, and prediluted standards. Simply dilute the reagent using the buffer provided, add your sample (any volume between 1 μL and 20 μL is acceptable), and read the concentration using the Qubit 3.0 Fluorometer (Figure 1). The assay is performed at room temperature, and the signal is stable for 3 hours.

All Qubit assays are performed using the same general protocol, which utilizes a simple mix-and-read format, with incubation times of only 2 minutes required for DNA and RNA assays or 15 minutes for the protein assay). The Qubit 3.0 Fluorometer has on-instrument storage for up to 1,000 sample runs, and you can transfer data using the USB cable or USB drive.

In addition to our core assays for dsDNA, oligos, total RNA, microRNA, and protein, Ion Sphere™ Particle quality can be evaluated on the Qubit 3.0 Fluorometer using the Ion Sphere™ Quality Control Kit prior to performing a sequencing run on the Ion Personal Genome Machine™ (PGM™) Sequencer. The Qubit® quantitation assay workflow

PLEASE NOTE: All Qubit Assay Kits are compatible with the Qubit 3.0 and 2.0 Fluorometers and with the original Qubit (1.0) Fluorometer with the exception of the microRNA and MyQubit assays, which are only compatible with the Qubit 3.0 and 2.0 Fluorometers.

Qubit assays (read using the Qubit 3.0 Fluorometer) are ideal when you need to quantitate 1–20 samples.
For 20–2,000+ samples, use Quant-iT™ Assay kits and reagents for use with microplate readers for nucleic acids or protein quantitation.

Select a Qubit assay kit based on target molecule

The Qubit dsDNA BR (broad range) and HS (high sensitivity) Assay Kits are designed for easy and accurate DNA quantification. These assays are highly selective for double-stranded DNA (dsDNA) over RNA. Common contaminants, such as salts, free nucleotides, solvents, detergents, or protein are well tolerated in the assay.

To determine the purity of your sample, use a Qubit dsDNA Assay Kit together with a Qubit RNA Assay Kit. These measurements will give you a much better indication of sample purity than that produced by an A260/A280 measurement. To measure protein contamination in nucleic acid samples, simply assay 1 to 20 μL of the sample in the Qubit Protein Assay.

The Qubit RNA BR (broad range) and HS (high sensitivity) Assay Kits offer easy and accurate RNA quantification. The assay is highly selective for RNA over double-stranded DNA (dsDNA) and is accurate for initial sample concentrations from 1 ng/μL to 1 μg/μL, providing an assay range from 20–1,000 ng for the BR assay, and from 250 pg/μL to100 ng/μL, providing an assay range of 5–100 ng for the HS assay. Common contaminants, such as salts, free nucleotides, solvents, detergents, or protein are well tolerated in the assay.

To determine the purity of your sample, use a Qubit dsDNA Assay Kit together with a Qubit RNA Assay Kit. The Qubit microRNA Assay Kit is designed for easy and accurate quantification of small RNA (17–25 nt or bp) such as microRNA, even in the presence of common contaminants such as salts, free nucleotides, solvents, detergents, and protein. The assay is highly selective for small RNA over rRNA or large mRNA (>1,000 bp). We have been able to reproducibly quantify small RNA in pure samples at levels as low as 0.5 ng in the assay tube (following the protocol in the supplied user manual). The assay detects all types of small RNA, including microRNA and siRNA, both single stranded and double stranded. The assay has a dynamic range of 5 ng/mL to 500 ng/mL (1–500 ng) in the core assay range and can detect as little as 2.5 ng/mL (0.5 ng) in the extended assay range. The assay is accurate for initial sample concentrations from 0.05 ng/μL to 100 ng/μL.

The assay is performed at room temperature, and the signal is stable for 30 minutes. The Qubit Protein Assay Kits are designed to make protein quantitation easy and accurate. The assay is accurate for initial sample concentrations from 12.5 μg/mL to 5 mg/mL and exhibits low protein-to-protein variation. Common contaminants, such as reducing reagents (DTT, β-mercaptoethanol), salts, free nucleotides, amino acids, solvents, and DNA (but not detergents) are well tolerated in the assay; some very slight modifications for the procedure are required for other contaminants.

Operating in Fluorometer mode

You have the option of measuring assay samples in the Qubit 3.0 Fluorometer in Fluorometer mode. In Fluorometer mode, you manually select the excitation source (blue LED or red LED), and the instrument generates and displays raw fluorescence (RFU) values for each sample.

Extending the capabilities of your Qubit Fluorometer

You can add new assays to your Qubit Fluorometer via simple download and transfer of .qbt files using a USB cable or USB drive.

Ion Sphere Assay

The Ion Sphere™ assay on the Qubit 3.0 Fluorometer enables quick and easy assessment of Ion Sphere Particles (ISPs) prior to performing a sequencing run on the Ion Personal Genome Machine™ (PGM™) Sequencer using the Ion Sphere Quality Control Kit. All ISPs bind a complimentary, green-fluorescent Alexa Fluor™ 488 dye-labeled oligonucleotide, whereas only those beads that are template-positive bind a complementary red-fluorescent Alexa Fluor 647 dye-labeled oligonucleotide. The Qubit 3.0 Fluorometer measures the fluorescence emission of each dye conjugate, and an Excel™ file template is used to calculate the percentage of template-positive ISPs from the ratio of the Alexa Fluor 647 dye signal to the Alexa Fluor 488 dye signal. This quality-control step using the Qubit Fluorometer helps determine the likelihood of a successful sequencing run, ultimately saving time and expense during subsequent sequencing.

The Ion Sphere™ Particle (ISP) assay principle using Alexa Fluor™ dyes. The B nucleic acid sequence is present in both the ISP primer and the template and is used to drive the amplification of the template onto the bead; the A sequence is only present in the template. The ratio of fluorescence signals from Alexa Fluor 647 and Alexa Fluor 488 dye–labeled oligonucleotides is used to determine the relative amount of template-positive particles.
