Carbodiimide crosslinking reagents, DCC and EDC (EDAC), are used for the conjugation of carboxyl groups (glutamate, aspartate, C-termini) to primary amines (lysine, N-termini) and N-hydroxysuccinimide (NHS) for stable activation of carboxylates for amine-conjugation.
A variety of carboxyl-to-amine crosslinkers are available:
- DCC is water-insoluble carbodiimide-based crosslinker that activates carboxyl groups for spontaneous peptide-bond formation with primary amines.
- EDC is a water-soluble carbodiimide-based crosslinker that activates carboxyl groups for spontaneous reaction with primary amines, enabling peptide immobilization and hapten-carrier protein conjugation.
- NHS- and sulfo-NHS crosslinkers are chemical modification reagents for converting carboxyl groups to amine-reactive NHS esters for bioconjugation, crosslinking, labeling, and immobilization.
