FAQs: Substrate Identification Assays

Why does the phosphorylation of some autophosphorylating proteins go down at higher kinase concentrations?
The assays are performed at a limiting ATP concentration. At the highest kinase concentration, ATP bound by kinase is depleted from the assay solution. The result of this effect is lower autophosphorylation, and a lower signal on the arrays.

How specific are the substrate phosphorylations identified in this experiment?
Comparing phosphorylation patterns from additional kinases will provide information regarding the specificity of the phosphorylation events identified in this project. To investigate this further, you can select or submit additional kinases for substrate identification.

How can I characterize substrate phosphorylations by the kinase in the presence of a small molecule, peptide, or protein that may modify kinase phosphorylation activity?
Repeating the assays in the presence of these modifiers may provide additional information regarding the mechanism of the kinases.
