Investigate cAMP Influence on JAK/STAT Pro-Inflammatory Signaling Pathways

  • Broad dynamic range—0.06 to 6,000 pmol (4 to 5 logs)
  • Flexible—can be used in manual or automated, high-throughput formats
  • Simple—sample dilution not necessary

Cyclic adenosine monophosphate (cAMP) is an important second messenger in many signal transduction pathways linking activation of cell surface membrane receptors to intracellular responses and, ultimately, to changes in gene expression. This competitive heterogeneous immunoassay measures cAMP directly in cell lysates at levels as low as 60 fmol to 6 µmol, without requiring any sample dilution. Detection is typically complete within 90 minutes, utilizes chemiluminescence, and is amenable to use in automated high-throughput screening applications.

The sensitivity and simplicity of the cAMP-Screen Direct® assay will enable the researcher to measure small changes in cAMP levels in the investigation of JAK/STAT pro-inflammatory signaling pathways.

New Antibodies to STAT Family Proteins

  • ABfinity™ rabbit recombinant antibodies for sensitive detection of STAT proteins
  • Enables consistent performance for your assays
  • Consistent product from lot to lot

ABfinity™ recombinant rabbit antibodies are produced from specific recombinant clones to enable consistent antibody performance over time. ABfinity™ antibodies are available against many targets, including several STAT isoforms (gene IDs: 6772, 6773, 6774, 6776, 6778).

The STAT (signal transducer and activator of transcription) family proteins regulate gene expression in cellular growth, survival, and differentiation. There are seven STAT family members: STAT1, 2, 3, 4, 5a, 5b, and 6. The STAT transcription factors are activated by Janus kinase (JAK), and this pathway, particularly the persistent phosphorylation of STAT3 and 5, may be correlated with oncogenic activity, making these proteins potential targets for drug treatments. Antibodies against the various STAT proteins and their active phosphorylated states are useful tools for investigating the regulation of STAT proteins as well as understanding the biological consequences of STAT phosphorylation.

Indirect ELISA was performed using dilutions of STAT3 Figure 1. Indirect ELISA was performed using dilutions of STAT3 [pY705] ABfinity™ Recombinant Rabbit Monoclonal Antibody to detect endogenous STAT3 protein in insulin-treated HeLa cells (cells were coated onto the plate). A nonlinear regression analysis was performed (4 PL), and LOD and LOQ for the antibody were determined. If possible to edit the x-axis, please show it as “log Concentration (µg/mL)”.

New Gibco® Cytokines that Stimulate the JAK/STAT Pathway

Gibco® Recombinant PDGF products offer:

  • High biological activity
  • High purity
  • Proven compatibility with Gibco® media

STAT proteins are a family of cytoplasmic signaling proteins involved in signal transduction of several cytokines and growth factors. In response to the cytokine or growth factor binding to cell surface receptors, STAT proteins become activated by tyrosine phosphorylation. Phosphorylated STAT proteins subsequently move to the cell nucleus, where they activate transcription.

In the past year, we have released several Gibco® recombinant proteins known to activate the JAK/STAT pathway. In April we launched human expressed IL-23, which interacts with the IL-23 receptor and the IL-12 receptor beta to activate JAK2 and STAT2. In July we launched human expressed IL-9, IL-28A, IL-28B, and GH. IL-9 interacts with the IL-9 receptor to activate JAK1 and 3, and STAT 1, 3, and 5. IL-28A and IL-28B (in combination with IL-29) interact with the IL-28 receptor and the IL-10 receptor to activate JAK1 and STAT1, 2, 3, 4, and 5. Human growth hormone (HGH) interacts with the growth hormone receptor, activating JAK2 and STAT3 and 5.

Gibco® cytokines and growth factors are high-purity recombinant proteins with high bioactivity. Gibco® growth factors are of the highest quality because each protein is analyzed for purity along with structural homogeneity to help ensure that a biologically active protein is produced. And Gibco® cytokines and growth factors have been bioassayed with Gibco® media. Get your cytokines and growth factors from the media company you use and trust.

Introducing the new FLoid™ Cell Imaging Station

FLoid™ Cell Imaging Station

  • Simple—collect images quickly with the intuitive user interface
  • Practical—print cell images and place them in your notebook
  • Accessible—capture fluorescent cell images at your bench, not in the darkroom

Designed with input from fluorescence microscopy users, the FLoid™ Cell Imaging Station captures high-quality fluorescence images right at your benchtop, with an interface so simple that even novice users can collect data with just a few clicks of the mouse.

The FLoid™ Cell Imaging Station allows you to image fluorescently labeled cells on slides, multiwell tissue culture dishes, flasks, etc., for routine analysis or to confirm staining before you move on to expensive, difficult-to-use confocal microscopes. This instrument is ideal for researchers studying the JAK/STAT pathway, who rely on fluorescence imaging to identify changes in phosphorylation states, cellular localization, and expression of proteins of interest following stimulation.

Screen Drug Inhibitors of STAT3 Phosphorylation in Cancer Research

  • Easy-to-run ELISA, 4 hours of incubation time
  • Convenient precoated, removable 8-well strips
  • Quantitative, sensitive results

JAK/STAT regulates cell proliferation, differentiation, and apoptosis. Various cytokines may initiate JAK activation, which then causes downstream phosphorylation of STAT proteins that ultimately translocate into the nucleus to control transcription of genes. Identifying inhibitors of the JAK/STAT protein family is a big focus in cancer research.

The STAT3 [pY705] ELISA Kit is a great screening tool for drug inhibitors; it enables easy, reliable measurement of phosphorylated STAT3 in cell lysates. The assay can be performed in a 96-well plate format and quantitative results obtained within a day. Convenient strip-well plates also provide flexibility to run as few samples as needed without wasting material.
