Cyclic adenosine monophosphate (cAMP) is an important second messenger in many signal transduction pathways linking activation of cell surface membrane receptors to intracellular responses and, ultimately, to changes in gene expression. This competitive heterogeneous immunoassay measures cAMP directly in cell lysates at levels as low as 60 fmol to 6 µmol, without requiring any sample dilution. Detection is typically complete within 90 minutes, utilizes chemiluminescence, and is amenable to use in automated high-throughput screening applications.
The sensitivity and simplicity of the cAMP-Screen Direct® assay will enable the researcher to measure small changes in cAMP levels in the investigation of JAK/STAT pro-inflammatory signaling pathways.

The STAT (signal transducer and activator of transcription) family proteins regulate gene expression in cellular growth, survival, and differentiation. There are seven STAT family members: STAT1, 2, 3, 4, 5a, 5b, and 6. The STAT transcription factors are activated by Janus kinase (JAK), and this pathway, particularly the persistent phosphorylation of STAT3 and 5, may be correlated with oncogenic activity, making these proteins potential targets for drug treatments. Antibodies against the various STAT proteins and their active phosphorylated states are useful tools for investigating the regulation of STAT proteins as well as understanding the biological consequences of STAT phosphorylation.

STAT proteins are a family of cytoplasmic signaling proteins involved in signal transduction of several cytokines and growth factors. In response to the cytokine or growth factor binding to cell surface receptors, STAT proteins become activated by tyrosine phosphorylation. Phosphorylated STAT proteins subsequently move to the cell nucleus, where they activate transcription.
In the past year, we have released several Gibco® recombinant proteins known to activate the JAK/STAT pathway. In April we launched human expressed IL-23, which interacts with the IL-23 receptor and the IL-12 receptor beta to activate JAK2 and STAT2. In July we launched human expressed IL-9, IL-28A, IL-28B, and GH. IL-9 interacts with the IL-9 receptor to activate JAK1 and 3, and STAT 1, 3, and 5. IL-28A and IL-28B (in combination with IL-29) interact with the IL-28 receptor and the IL-10 receptor to activate JAK1 and STAT1, 2, 3, 4, and 5. Human growth hormone (HGH) interacts with the growth hormone receptor, activating JAK2 and STAT3 and 5.

JAK/STAT regulates cell proliferation, differentiation, and apoptosis. Various cytokines may initiate JAK activation, which then causes downstream phosphorylation of STAT proteins that ultimately translocate into the nucleus to control transcription of genes. Identifying inhibitors of the JAK/STAT protein family is a big focus in cancer research.
